CHROMagar is a ready-to-use medium that needs to be rehydrated before use. To prepare CHROMagar medium, dissolve the powder in distilled or deionized water, heat until boiling, cool to 45-50°C, pour into sterile Petri dishes, and allow to solidify at room temperature. Store the plates in a sealed plastic bag at 2-8°C until use.
CHROMagar allows the identification of bacteria based on colony color and morphology. Different bacteria produce different colony colors and characteristics on CHROMagar. However, atypical colony colors or morphology can occur, so CHROMagar should be confirmed by other methods for bacterial identification.
CHROMagar has several advantages over conventional media and finds applications in clinical microbiology, food microbiology, water microbiology, and environmental microbiology. It enables the simultaneous detection and identification of multiple organisms, enhances contrast, detects organisms that may be missed on conventional media, and facilitates enumeration of organisms.
